رنگ آمیزی با لاکتوفنل کاتن بلو:
این رنگ برای تهیه گسترش مرطوب قارچها مناسب می باشد.
روش رنگ آمیزی :
روی یک لام تمیز یک قطره از محلول لاکتوفنل کاتن بلو ریخته بعد با استفاده از آنس استریل کمی از پرگنه قارچ را برداشته و با دقت روی لام قرار دهید . نمونه نباید زیاد باشد چون مشاهده میکروسکوپی آن مشکل می شود . نکته قابل توجه اینکه هنگام برداشت پرگنه ، نوک آنس را زیاد حرکت ندهید چون اینکار باعث بهم ریختن بافت قارچ می شود . همچنین بعد از اضافه کردن پرگنه روی محلول رنگی آنرا هم نزنید چون بافت قارچ پاره می شود و بررسی آن مطلوب نیست . بعد یک لامل روی آن گذاشته با میکروسکوپ با بزرگنمایی 10 و 40 بررسی کنید.
برای بررسی با بزرگنمایی 100 باید بعد از قرار دادن لامل چند ضربه آهسته به آن زده و بعد با استفاده از روغن سدر سطح لامل را آغشته کرده و بررسی کنید.Gram-color staining kit enables distinctions to be made between gram-positive and gram-negative bacteria.
