Urea Broth Base
Use and description: This is a liquid Christensen’s medium formulation without agar which allows inoculation by Pasteur pipette. Suitable for the differentiation of urease-producing members of the Salmonella and Shigella groups. Rapid growth ensued and quick positive reaction is achievable. Urease-positive bacteria turn the medium pink.
Composition per liter: Sodium chloride……………………………..5.000 g Disodium hydrogen phosphate……………...1.200 g Peptone……………………………………...1.000 g Glucose……………………………………...1.000 g Potassium dihydrogen phosphate…………...0.800 g Phenol red…………………………………...0.004 g
Final pH of the ready to use medium: 6.8 ± 0.2
Medium preparation: Add 9 grams of dehydrated culture medium to 950 mL of distilled water and bring volume to 95 ml. Mix thoroughly. Sterilise by autoclaveing at 115oC for 20 minutes. Cool to 50 oC and aseptically add 50 ml of sterile Urea solution. Mix thoroughly. Aseptically distribute into sterile tubes or flasks.
Quality specification: Dehydrated medium: homogeneous, creamy, fine powder. Ready to use medium: clear, yellow broth.
Microbiological response:
Storage: Dehydrated medium should be stored between 10 to 25oC. Once opened, place the container in a dark, dry place. The dehydrated medium should not be used if there is any lump or the color has changed from the original.
