محیط کشت بریلیانت گرین آگار Briliant green agar
محیطی است انتخابی و برای جداسازی گونه های سالمونلا بکار می رود. این محیط از رشد بسیاری از باکتریها ی روده ای (انتروباکتریاسه) به جز سالمونلا جلوگیری می نماید. در صورتی که Ecoli ،کلبسیلا و پروتئوس و سایر انترو باکتریاسه ها رشد نمایند ، به دلیل اینکه قند لاکتوز ویا سوکروز ویا هر دو را تخمیر می نمایند و تولید اسید می کنند، در مجاورت معرف رنگی فنل رد به رنگ زرد در می آیند .در صورتی که باکتری های لاکتوز منفی مانند سالمونلا رشد نمایند ، به دلیل عدم تخمیر قند در مجاورت معرف ، محیط به رنگ ارغوانی در می آید.Description of MM0167

Brilliant Green Agar (Modified) (BGA) Use and description: Medium for the selective isolation of salmonellae other than Salmonella typhi. The modification was decrease Brillant green concentration for higher recovery rate. Salmonella other than S. typhi appear as red/pink/white colonies surrounded by a zone of red in the agar, indicating nonlactose/sucrose fermentation. Lactose and/or sucrose fermenting bacteria appear as green colonies

Final pH of the ready to use medium: 6.9 ± 0.2
Medium preparation: Add 52.0 grams of dehydrated culture medium to 1 liter of distilled water. Heat gently with occasional agitation and bring just to the boil to dissolve the medium completely. Addition of 1.0 gram of sodium sulfacetamide and 250.0 milligrams sodium mandelate enhances inhibition of contaminating microorganisms. DO NOT AUTOCLAVE. Pour into sterile Petri dishes.
Quality specifications: Dehydrated medium: homogeneous, pinkish fine powder. Ready to use medium: orange-brown agar, very slightly opalescent.
Microbiological response:
Storage: Dehydrated medium should be stored between 10 to 25oC. Once opened, place the container in a dark, dry place. The dehydrated medium should not be used if there is any lump or the color has changed from the original. |
