Brain Heart Infusion Broth (BHI Broth)
Use and description: Brain Heart Infusion Broth is used for the cultivation of a wide variety of fastidious microorganisms, including streptococci, pneumococci and meningococci and further bacteria, yeasts and moulds, especially fastidious species. The addition of 0.1% agar can be used to lower oxygen tension which improves the growth of anaerobic organisms. It contains no bovine component to minimize BSE contamination risk.
Composition per liter: Proteose peptone………………………………..10.0 g Brain heart infusion solid (pork).……………….17.5 g Dextrose………………………………………….2.0 g Sodium chloride………………………………….5.0 g Disodium hydrogen phosphate…………………..2.5 g
Final pH of the ready to use medium: 7.4 ± 0.2
Medium preparation: Add 37.0 grams of dehydrated culture medium to 1 liter of distilled water. Heat with repeated stirring and boil for one minute to dissolve completely. Distribute into tubes of flasks. Autoclave for 15 min at 121oC. Overheating will cause caramelisation and darkening of the medium.
Quality specifications: Dehydrated medium: homogeneous, light beige fine powder. Ready to use medium: clear and light to medium straw.
Microbiological response at 37 °C, for 18-48 hours:
Storage: Dehydrated medium should be stored between 10 to 25oC. Once opened, place the container in a dark, dry place. The dehydrated medium should not be used if there is any lump or the color has changed from the original. |
