TCBS Agar (Thiosulphate Citrate Bile Salt Sucrose Agar)
Use and description: T.C.B.S. is designed for the selective isolation of Vibrio cholerae and Vibrio parahaemolyticus. The high concentrations of thiosulphate and sodium citrate inhibit the growth of enterobacteria at least for 24 hours. Ox bile and sodium cholate slow the growth of enterococci and inhibit Gram-positive bacteria. Hydrogen sulphide production is visualized by blackening of media. All Vibrio are H2S-negative and sucrose fermenters Vibrio spp. appear as yellow colonies becouse of the acid production which turns the indicator from green to yellow.

Final pH of the ready to use medium: 8.6 ± 0.2
Medium preparation: Add 87.0 grams of dehydrated culture medium to 1 liter of distilled water, mix until evenly dispersed. Heat with repeated stirring and boil for one minute. Poured plates can be prepared after the medium has been cooled to 45-50oC. DO NOT AUTOCLAVE THIS MEDIUM!
Quality specification: Dehydrated medium: homogeneous, beige to greenish-beige, fine powder. Ready to use medium: clear to trace hazy and forest green.
Microbiological response:
Storage: Dehydrated medium should be stored between 10 to 25oC. Once opened, place the container in a dark, dry place. The dehydrated medium should not be used if there is any lump or the color has changed from the original.
