TSB U.S.P (Tryptone Soy Broth)
Use and description: The general purpose broth for growing a wide range of bacteria and fungi. The medium is recommended by the United States Pharmacopeia for the sterility testing. It can be used for aerobic or anaerobic cultivation with the addition of a small amount of agar (0,1-0,2%).
Composition per liter: Sodium chloride………………………………5.0 g Soy peptone…………………………………...3.0 g Tryptone……………………………………..17.0 g Dipotassium hydrogen phosphate…………….2.5 g Glucose.………………………………………2.5 g
Final pH of the ready to use medium: 7.3 ± 0.2
Medium preparation: Add 30 grams of dehydrated culture medium to 1 liter distilled water. Heat with repeated stirring and boil for one minute to dissolve completely. Distribute and autoclave for 15 min at 121oC.
Quality specification: Dehydrated medium: homogeneous, light-beige, fine powder. Ready to use medium: clear and very light amber.
Microbiological response:
Storage: Dehydrated medium should be stored between 10 to 25oC. Once opened, place the container in a dark, dry place. The dehydrated medium should not be used if there is any lump or the color has changed from the original. |
