Yeast Extract Agar
Use and description:
This medium is used for the plate count of heterotrophic microorganisms in water and dairy products.
Composition per liter:
Peptone……………………………………5.0 g
Yeast extract………………………………3.0 g
Agar……………………………………...14.0 g
Final pH of the ready to use medium: 7.2 ± 0.2
Medium preparation:
Add 22 grams of dehydrated culture medium to 1 liter of distilled water. Heat gently and bring to boiling to dissolve completely. Mix well and dispense into containers. Sterilise for 15 min at 121oC.
Quality specification:
Dehydrated medium: homogeneous, beige fine powder.
Ready to use medium: clear gel, pale straw-coloured

Storage:
Dehydrated medium should be stored between 10 to 25oC. Once opened, place the container in a dark, dry place. The dehydrated medium should not be used if there is any lump or the color has changed from the original.
